My laboratory is working on the development of small molecule inhibitors of transcription factors that are drivers in cancer. We recently developed a small molecule inhibitor targeting the CBFB-SMMHC fusion protein that is a driver in ~8% of acute myeloid leukemia (AML) cases ((Illendula et al. Science 347(6223):779-84 (2015); Pulikkan et al. Cell 174(1):172-186 (2018)). This compound has been licensed to Systems Oncology and is being developed for clinical evaluation. We have also developed small molecule inhibitors targeting the wildtype CBFB-RUNX interaction (Illendula et al. EBioMedicine 8:117-131 (2016)) and shown these are effective in a number of cancer contexts including leukemia (Illendula et al. EBioMedicine 8:117-131 (2016), Choi et al. Blood 130(15):1722-1733 (2017)), multiple myeloma (Zhou et al. Leukemia, in press), triple negative breast cancer (TNBC) (Illendula et al. EBioMedicine 8:117-131 (2016)), and ovarian cancer (Carlton et al. Gynecol. Oncology 149(2):350-360 (2018)).
