Acute lymphoblastic leukemia (ALL) kills more children than any other cancer. Our goal is use cutting-edge transcriptional genomic and proteomic technologies and sophisticated preclinical animal and human models to identify and elucidate the network of transcriptional regulators, genes and regulatory elements of ALL and regenerating T-cells in order to develop novel targeted anti-leukemic therapies that are more effective and less toxic than current medicines. We will also apply this knowledge towards developing medicines that promote recovery of T-cell immunity after cytoreductive therapy.

Specific methodologies:
-Leukemogenesis and T-cell development assays (flow cytometry sorting and analysis; histology; bone marrow transplantation; intrathymic injections; retroviral transduction and transplantation; in vitro stromal cell culture; transplantation in NSG mice)
-Transcriptional genomics (ChIP-Seq/ATAC/4C-Seq library preparation; analyses of RNA/ChIP/ATAC-seq datasets using computational pipelines; bioinformatics)
-Transcription factor/enhancer functional screens and assays (CRISPRi; ChIP; MPRAs)
-Protein-protein interaction biochemistry (mass spectrometry; BIOID; NMR; co-IP; reverse-ChIP)
Model systems:
- Genetically engineered mouse models
- Human leukemia patient-derived xenograft models
- Human and mouse leukemia cell lines
Specific responsibilities:
-Design, perform, analyze, and interpret experiments with training and supervision from PI and other lab members in line with skill set and with goal of independence
-Work as a team with other lab members (PI, bioinformaticist, postdocs, PhD students, and the lab manager/work-study students)
-Collaborate with members of other labs
-Write manuscripts for publication
-Present posters and talks at research conferences
