Job Description
We are looking for an expert post-doc to join the NeuroGlio team within the Cancer and Brain Genomics INSERM unit and the Normandy Institute of Precision Medicine, to work on the elucidation of the molecular and metabolic signatures of the glioblastoma GlioVascular Unit (INCA PAIR Brain Tumor project - GlioVasc Project). The global aim of the whole program is to elucidate the key features of the gliovascular unit and its pivotal role in glioblastoma (GB) recurrence and treatment resistance. We propose to explore the dynamic trajectories of the GB and stromal cells within the gliovascular unit from human GB samples during the Stupp protocol. Our NeuroGlio team will map the gliovascular unit heterogeneity phenotypically (organoids) and at the single cell level in GB patient samples to identify gene targets to validate in GB resection preclinical models.

Team NeuroGlio is expert in Neuroscience and Neuro-oncology in a highly complementary environment of the Cancer and Brain Genomic (CBG) laboratory exploring genetic predisposition of tumors and neuronal and neurodevelopmental pathologies. The main aims of the team are to identify new therapeutic targets when glioblastoma grow in its neuroimmune microenvironment, and to evaluate the brain sequelae related to cancer and cancer treatments, between therapeutic efficacy and preservation of brain functions. NeuroGlio expertise resides in molecular and cellular biology of migration and invasion of glioma cells, in neuroscience features (electrophysiology/optogenetics) guiding glioma growth, in behavioral mouse models with cancer, after microsurgery, and during conventional treatments or immunotherapy. Clinical protocol and methodologies to establish GB tumoroïds from patients and glio-cerebroids from human iPS cells are currently leveraged.
Skills and approaches: Establishment glioblastoma tumoroïds and vessels isolation from GB patient samples, from the surgical block to the laboratory (maintenance, optimization of intra-tumor heterogenous cell composition and dynamic imaging). Patient derived cell lines will also be systematically generated. From selected patient samples, expertise in spatial single cell RNA-seq and spatial metabolomics from the GB gliovascular unit is required. From tumoroids and GB xenografted mice, key molecular targets of the gliovascular niche will then be tested. All facilities are available within the Institute for Research and Innovation in Biomedicine of Rouen.
Candidat profile: The ideal candidate should have excellent teamwork skills in 2D/3D cell culture from tissue and in organoids maintenance, microscopy and 3D imaging/use of the Imaris software, skills in conducting single cell RNAseq experiments from tissue samples. Expertise in in vivo preclinical models of GB should be a plus. He/she would be fluent in both spoken and written English.
